The annual event will start in Bowral on Sunday morning, with cyclists competing in 85km, 120km and 150km rides across the region.
Bong Bong Street in Bowral will be closed in both directions between Banyette Street and Merrigang Street between 4am and 9am.
Local road closures and detours will also be in place at various times from 4am to 4.30pm through Bowral, Berrima, Mittagong, Bundanoon, Fitzroy Falls, Robertson and Kangaloon.
Motorists are reminded to leave a minimum distance of 1.5 metres when overtaking cyclists on roads with speed limits above 60km/h and one metre on roads with speed limits of 60km/h or lower.
